All themes are compatible with GD out of the box.
Sometimes few classes must be edited in theme compatibility settings to adjust GD templates width.
Before it was done via a compatibility plugin:

We don’t have detailed documentation for a similar task.

locations are created when submitting listings if they are not already in your database. No need to create them in advance.
The multi location settings you had are wrong. Enabling default city will only allow to create listings within the default city. I enable multi city. Nobody will be able to add a listing in a city that is outside your selected regions.

GeoDirectory is a WordPress plugin, so yes it can only work within WordPress.
If you don’t have knowledge of PHP HTML and CSS, it’ll be very hard to achieve anything decent. The best thing to do would be to do your design in illustrator or whatever you use and hire a developer to turn your design into a theme compatible with GeoDirectory.

listings must have a physical address and they must be assigned to a city. You can’t assign a listing to a region.

I don’t think there is anyway to do something like that.
Those settings are to improve SEO and adding the same description to all locations just adding the location as a variable won’t help with SEO at all.
If that’s what you want to do anyway, you should use a text widget and the location shortcodes as variables.
